More Information from Your Sample
The PowerPlex® 35GY System is the first 8-dye STR multiplex and allows co-amplification and fluorescent detection of the 20 CODIS core loci plus Amelogenin and DYS391 for sex determination. The inclusion of two additional dye channels relative to previous STR multiplex kits provides improved and more informative analyses for even your most challenging DNA samples.
- Fifteen mini-STRs improve results with degraded samples.
- System contains the shortest SE33 amplicon of any STR chemistry kit available.
- Penta D, Penta E and SE33 loci allow wider database searching.

The Most Loci in Any STR Multiplex
The power of 8 dye channels. Shifting larger autosomal loci in the PowerPlex® Fusion 6C System into two additional dye channels in the PowerPlex® 35GY System frees up space for Y-STR loci to be included, providing more information from a sample and improving analysis.
One Kit for All Your Sample Types
Designed for amplification of extracted DNA and direct amplification of single-source reference samples, the PowerPlex® 35GY System enables laboratories to process all sample types with one amplification kit. The PowerPlex® 35GY System is optimized to work with 1ng of extracted DNA in a 25μl reaction volume and is compatible with the Spectrum CE System and Spectrum Compact CE System.
2800M Control DNA–Spectrum CE System
Amplifying extracted DNA with the PowerPlex® 35GY System. The 2800M Control DNA (1ng) was amplified using the PowerPlex® 35GY System. Amplification products were mixed with CCO Internal Lane Standard 500 and separated on the Spectrum CE System. The electropherogram was generated using GeneMarker® HID Software for Spectrum CE Systems. Dye names are shown on the right.
2800M Control DNA–Spectrum Compact CE System
Amplifying Extracted DNA with the PowerPlex® 35GY System. The 2800M Control DNA (1ng) was amplified using the PowerPlex® 35GY System. Amplification products were mixed with CCO Internal Lane Standard 500 and separated on the Spectrum Compact CE System. The electropherogram was generated using GeneMarker® HID Software for Spectrum CE Systems.
As with our 5-dye and 6-dye PowerPlex® Systems, the PowerPlex® 35GY System also supports reduced-volume direct amplification reactions. In one example, DNA from a buccal swab in SwabSolution™ Reagent was amplified for 25 cycles in a 12.5μl reaction volume. Amplified samples were separated on the Spectrum CE System.
Buccal FTA® Punch
Direct amplification with the PowerPlex® 35GY System. An FTA® punch of buccal cells was direct-amplified using the PowerPlex® 35GY System. Amplification products were separated on the Spectrum CE System and analyzed using GeneMarker® HID Software for Spectrum CE Systems.
Quality Indicators Ease Data Interpretation
To help you better interpret the quality of your STR results, the PowerPlex® 35GY System is the first PowerPlex® system to include two Quality Indicators, QIS and QIL. The Quality Indicators take the guesswork out of determining if amplification was successful by confirming if DNA is present or if the sample amplification was inhibited. Knowing whether a sample was degraded or inhibited lets you decide whether to repeat sample analysis.
At-a-Glance Quality Analysis
Quality indicators for the PowerPlex® 35GY System. Balanced QIS and QIL peaks indicate a successful STR profile (Panel A). An STR profile with balanced QIS and QIL peaks but no STR peaks indicates that no template DNA was amplified (Panel B). A sloping STR profile with balanced QIS and QIL peaks signals degraded sample (Panel C). A sloping STR profile with a low or missing QIL peak suggests the sample is inhibited (Panel D).
More Successful Mixture Deconvolution
Mixed casework samples often contain PCR inhibitors, and the minor DNA component may be present at a very low level. The enhanced sensitivity and superior capability to overcome inhibition, as well as inclusion of 11 Y-STR loci in the PowerPlex® 35GY System, enable laboratories to interpret mixed samples more confidently than previously possible.

Better mixture analysis with more Y-STRs in the PowerPlex® 35GY System. Extracted DNA (1ng) from four-person mixtures was amplified using the PowerPlex® 35GY System. Amplification products were separated on the Spectrum CE System and analyzed using GeneMarker® HID Software for Spectrum CE Systems.
Mini-STRs Improve Success with Difficult Samples
The environmental conditions encountered in forensic casework, mass disasters and missing persons scenarios can lead to severe degradation of human DNA samples. Such highly degraded samples often produce incomplete or no STR profiles due to larger loci that can “drop out”. The inclusion of more loci as mini-STRs in the PowerPlex® 35GY System results in better and more complete profiles, making this system an ideal option to process cold-case samples and newer, difficult sample types.
In the PowerPlex® 35GY System, there are eight common loci with amplicons that are at least 100bp smaller in size than when amplified using the PowerPlex® Fusion 6C System.

Multi-laboratory studies confirm that this advanced multiplex, which includes fifteen mini-STRs, consistently generates high-quality DNA profiles that are eligible for inclusion in NDIS.
